Kisspeptin (Kp-10) is a neuropeptide that binds to GPR54 receptors, exerting several functions mainly in the nervous and reproductive systems of the body. However, its effects and mechanisms of action on the skeletal system remain poorly understood. This study evaluated the effects of different concentrations of Kp-10 on in vitro osteogenic differentiation of multipotent mesenchymal stromal cells (MSCs) extracted from the bone marrow (BM) of adult Wistar rats. Two-month-old female rats were euthanized to extract BM from long bones to obtain MSCs. Four experimental groups were established in vitro: a control and Kp-10 at concentrations of 0.01, 0.05 and, 0.1 µg/mL. After induction of osteogenic differentiation, cell viability was evaluated using the 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl tetrazolium bromide (MTT) assay, alkaline phosphatase activity, collagen synthesis, percentage of area covered by MSCs/field and mineralized nodules/field, and immunocytochemistry of the GPR54 receptor tests. Furthermore, evaluation of gene transcripts for type I collagen, Runx-2, Bmp-2, bone sialoprotein, osteocalcin and osteopontin was performed using real-time RT-qPCR. It was observed that MSCs expressed GPR54 receptor to which Kp-10 binds during osteogenic differentiation, promoting a negative effect on osteogenic differentiation. This effect was observed at all the Kp-10 concentrations in a concentration-dependent manner, characterized by a decrease in the activity of alkaline phosphatase, collagen synthesis, mineralized nodules, and decreased expression of gene transcripts for type I collagen, osteocalcin, osteopontin, and Runx-2. Thus, Kp-10 inhibits in vitro osteogenic differentiation of MSCs extracted from the BM of adult Wistar rats.

Reflectance hyperspectroscopy is recognised for its potential to elucidate biochemical changes, thereby enhancing the understanding of plant biochemistry. This study used the UV-VIS-NIR-SWIR spectral range to identify the different biochemical constituents in Hibiscus and Geranium plants. Hyperspectral vegetation indices (HVIs), principal component analysis (PCA), and correlation matrices provided in-depth insights into spectral differences. Through the application of advanced algorithms-such as PLS, VIP, iPLS-VIP, GA, RF, and CARS-the most responsive wavelengths were discerned. PLSR models consistently achieved R2 values above 0.75, presenting noteworthy predictions of 0.86 for DPPH and 0.89 for lignin. The red-edge and SWIR bands displayed strong associations with pivotal plant pigments and structural molecules, thus expanding the perspectives on leaf spectral dynamics. These findings highlight the efficacy of spectroscopy coupled with multivariate analysis in evaluating the management of biochemical compounds. A technique was introduced to measure the photosynthetic pigments and structural compounds via hyperspectroscopy across UV-VIS-NIR-SWIR, underpinned by rapid multivariate PLSR. Collectively, our results underscore the burgeoning potential of hyperspectroscopy in precision agriculture. This indicates a promising paradigm shift in plant phenotyping and biochemical evaluation.

BACKGROUND: Chronic kidney disease, for which estimated glomerular filtration rate (eGFR) trajectories are early markers, is frequent in people living with HIV. SETTING: Identify eGFR trajectory patterns according to kidney function and assess associated factors over a 13-year follow-up period. METHODS: We evaluated longitudinal changes and its associated factors in eGFR of 3366 participants according to kidney function with a 2-level, linear, mixed model. RESULTS: Participants with initial kidney dysfunction experienced a slight eGFR increase, whereas others showed a slight decrease. A weak relationship was observed between baseline eGFR and its variation over time. Baseline eGFR was affected by age, CD4 + count, viral load, hypertension, hyperlipidemia, AIDS-defining illness and tenofovir (TDF) with integrase inhibitor (INSTI) or efavirenz. Significant factors for eGFR change included the following: in kidney dysfunction, CD4 + cell count of >350 cells per cubic millimeter and undetectable viral load increased eGFR, whereas TDF + protease inhibitor decreased eGFR; in mildly decreased kidney function, CD4 + cell count of >350 cells per cubic millimeter, AIDS-defining illness, and TDF + efavirenz increased eGFR, whereas age, hypertension, hyperlipidemia, and TDF + INSTI decreased eGFR; in normal kidney function, age, CD4 + cell count of > 350 cells per cubic millimeter, undetectable viral load, hypertension, hyperlipidemia, and TDF + INSTI decreased eGFR, whereas TDF + efavirenz increased eGFR (all P value for interaction < 0.05). CONCLUSION: Our findings suggest that eGFR trajectories varied widely between individuals in people living with HIV. In the lower eGFR group, virus-related factors were more relevant, whereas traditional risk factors for renal dysfunction were more prominent in the highest eGFR group.

Solanum glaucophyllum Desf. is a calcinogenic plant responsible for enzootic calcinosis that affects ruminants and causes alterations in bone and cartilaginous tissues, among others. It is believed that changes in cartilage tissue, with reduced bone growth, are due to hypercalcitoninism, caused by excess vitamin D. However, we hypothesized that S. glaucophyllum Desf. can act directly on chondrocytes and therefore, chondrocyte cultures from the epiphysis of the long bones of newborn rats were used as a model to elucidate the direct effects of S. glaucophyllum Desf. on bone growth. Plant samples were collected from Cañuelas, Argentina. An aliquot of the plant extract was used to quantify vitamin D (1,25(OH)2D3). The effects of the three concentrations of the plant extract were tested in cultures of chondrocytes extracted from the epiphyses of the long bones of 32 three-day-old Wistar rats. A control group (without extract), and three groups treated with different concentrations of plant extract were formed: group 1 (100 µL/L); group 2 (1 mL/L), and group 3 (5 mL/L), containing respectively 1 × 10-9 M, 1 × 10-8 M, and 5 × 10-8 M of 1,25(OH)2D3. After 7, 14, and 21 days of culture, MTT assay for cell viability, alkaline phosphatase activity, and quantification of the percentage of areas with glycosaminoglycans (GAG) stained with periodic acid-Schiff (PAS) were performed. On day 7, all chondrocytes in group 3, that is, those with the highest concentration of plant extract, died. On days 14 and 21, groups 1 and 2 showed a significant reduction in chondrocyte viability compared to the control. At 7, 14, and 21 days, groups 1 and 2 showed significantly lower alkaline phosphatase activity than the control. On day 21, group 2 showed a significant reduction in areas with PAS + GAGs. There were no significant differences between the groups in the expression of gene transcripts for Sox9, Col2, ColX, and aggrecan. The S. glaucophyllum Desf. extract directly affected growing rat chondrocytes by reducing viability, alkaline phosphatase activity, and GAG synthesis without altering the expression of gene transcripts for Sox9, Col2, ColX, and aggrecan, which may be one of the mechanisms by which there is a reduction in bone growth in animals intoxicated by the plant.

This study aimed to evaluate the perception of public places for the practice of physical activity and compare these differences according to health plan beneficiaries and non-beneficiaries. We used data from the 2019 National Health Interview Survey. The outcome was assessed through the following question, "Near your home, is there a public place (square, park, enclosed street, beach) to walk, exercise or play sport?". Descriptive analyses were performed using frequency and 95% confidence intervals (95%CI). The analyses were performed by comparing beneficiaries and non-beneficiaries of health plans and evaluating the results according to sociodemographic variables.The results include data from 20,230 beneficiaries (52.6% women) and 68,301 non-beneficiaries (54.6% women) of health plans, with most of the sample aged between 18 and 39.Overall, 52.1% of non-beneficiaries (95%CI: 51.0 - 53.2) and 67.4% of beneficiaries (95%CI: 65.8 - 68.9) reported having a place near their home for physical activity. We found a dose-effect trend regarding education level and places close to the residence to the practice of physical activity - the higher the level of education, the higher the perception of individuals who reported having adequate places for physical activity.Our findings showed that beneficiaries of health plans have more access to places near their homes to practice physical activity, as well as the most educated


O objetivo deste estudo foiavaliar a percepção de locais públicos para a prática de atividade física e com-parar essas diferenças segundo beneficiários e não beneficiários de planos de saúde. Foram utilizados dados da Pesquisa Nacional de Saúde de 2019. O desfecho foi avaliado por meio da seguinte questão: "Perto de sua casa, existe algum local público (praça, parque, rua fechada, praia) para caminhar, fazer exercícios ou praticar esportes?". Análises descritivas foram realizadas por meio de frequência e intervalo de confiança de 95% (IC95%). As análises foram realizadas comparando beneficiários e não beneficiários de planos de saúde e avaliando os resultados segundo variáveis sociodemográficas.Os resultados incluem dados de 20.230 beneficiários (52,6% mulheres) e 68.301 não-beneficiários (54,6% mulheres) de planos de saúde, ambos com maior parte da amostra com idades entre 18 e 39 anos. No geral, 52,1% dos não beneficiários (IC95%: 51,0 - 53,2) e 67,4% dos beneficiários (IC95%: 65,8 - 68,9) relataram ter um local próximo a sua casa para a prática de atividade física. Encontramos uma tendência dose-resposta em relação à escolaridade e locais próximos à residência para a prática de atividade física - quanto maior a escolaridade, maior a percepção de indivíduos que referiram ter locais adequados para a prática de atividade física.Nossos achados mostraram que os beneficiários de planos de saúde têm mais acesso a locais próximos a suas residências para praticar atividade física, assim como os mais escolarizados

Unlike the central nervous system, the peripheral one has the ability to regenerate itself after injury; however, this natural regeneration process is not always successful. In fact, even with some treatments, the prognosis is poor, and patients consequently suffer with the functional loss caused by injured nerves, generating several impacts on their quality of life. In the present review we aimed to address two strategies that may considerably potentiate peripheral nerve regeneration: stem cells and tissue engineering. In vitro studies have shown that pluripotent cells associated with neural scaffolds elaborated by tissue engineering can increase functional recovery, revascularization, remyelination, neurotrophin expression and reduce muscle atrophy. Although these results are very promising, it is important to note that there are some barriers to be circumvented: the host's immune response, the oncogenic properties attributed to stem cells and the duration of the pro-regenerative effects. After all, more studies are still needed to overcome the limitations of these treatments; those that address techniques for manipulating the lesion microenvironment combining different therapies seem to be the most promising and proactive ones.

OBJECTIVE: to identify the rate of reactive treponemal and non-treponemal tests in pregnant women during childbirth and to analyze the factors associated with this seroreactivity. METHOD: this is a cross-sectional, quantitative study with secondary sources of sociodemographic and clinical data on 2,626 pregnant women treated at a public maternity hospital in the interior of São Paulo, in 2020. For statistical analysis, Fisher's exact test, Mann-Whitney test and the logistic regression model were used. A difference of p < 0.05 was considered statistically significant. RESULTS: the rate of seropositivity for syphilis among pregnant women in this series was 2.74%. Among the groups with positive and non-reactive tests, marital status, occupation, place of residence and use of licit drugs indicated significant differences, but, in the final model, only unmarried marital status was associated with reactive tests (Odds Ratio: 0.169; Confidence Interval: 0.04-0.72; and p: 0.016). CONCLUSION: in this study, unmarried marital status was the only independent factor associated with seroreactivity for syphilis. Therefore, it is necessary to create strategies aimed at women in this condition, potentially reducing the rate of congenital syphilis.

OBJECTIVES: to analyze the evidence available in literature on factors associated with inadequate treatment of syphilis in pregnant women. METHODS: an integrative review, carried out in the LILACS, CINAHL, Web of Science, Scopus, PubMed and EMBASE databases, with controlled descriptors therapeutic and prenatal syphilis. RESULTS: nine publications composed the interpretative analysis, in which low education, income and maternal age, temporary lack of medication and HIV infection were associated with inadequate treatment of syphilis during pregnancy, in addition to delay or absence of prenatal care and receiving the 1st dose of penicillin, lack of tests or treatment less than 30 days before childbirth, and partners' low compliance with treatment. FINAL CONSIDERATIONS: among the main factors associated with inadequate treatment, clinical and sociodemographic aspects stand out, as well as failures in drug dispensing, prescription and monitoring of treatment of pregnant women and their partners by the health system.

Herein, we aimed to evaluate cultures of femoral chondrocytes from offspring of rats with intrauterine growth restriction (IUGR) induced by maternal hyperthyroidism. Fourteen adult female Wistar rats were divided into two groups, a control group and a group treated with daily L-thyroxine administration using an orogastric tube (50 µg/animal/day) during pregnancy. Three days after birth, the offspring were euthanized for chondrocyte extraction. At 7, 14, and 21 days, viability and alkaline-phosphatase (ALP) activity were assessed using the MTT assay and BCIP/NBT method, respectively, in a 2D culture. Pellets (3D cultures) were stained with periodic acid Schiff (PAS) to assess the morphology and percentage of PAS+ areas. The gene transcripts for Col2, Col10, Acan, Sox9, and Runx2 were evaluated by qRT-PCR. The MTT and ALP-assay results showed no significant differences between the groups. Maternal hyperthyroidism did not alter the chondrocyte morphology, but significantly reduced the percentage of PAS+ areas, decreased the expression of the gene transcripts of Col2 and Acan, and increased Sox9 expression. Maternal hyperthyroidism in rats alters the composition and gene expression of the matrix produced by chondrocytes from offspring with IUGR. This may be one of the mechanisms through which excess maternal thyroid hormones reduce offspring bone growth.

Tobacco genetic improvement programs, as well as the tobacco industry, require techniques that allow the estimation of its attributes in a fast and cheap way. The use of remote sensing through visible, near infrared and short-wave spectroscopy (Vis-NIR-SWIR) has been studied aiming to meet such demand. Thus, the aim of this work was to evaluate the use of Vis-NIR-SWIR spectroradiometer as a rapid tool to estimate alkaloids, sugars and yield of tobacco varieties. For that purpose, a study was carried out in a greenhouse with plants grown in pots (18 dm-3) containing nutrient solutions. The experimental design was completely randomized, with 30 treatments (tobacco varieties) and 10 repetitions. Tobacco leaf reflectance was collected at 13, 34 and 68 days after transplantation (DAT) with a plant-probe device connected to the spectroradiometer by an optical fiber. Subsequently, leaf analysis of alkaloids, sugars and yield were performed, and such attributes were estimated by using the Partial Least Squares Regression (PLSR), combined with the following pre-processing (PP) techniques: multiplicative scatter correction (MSC), Savitzky-Golay (SG) and standard normal variate (SNV). The results showed presence of typical inflections of chemical and structural components of the plants, which allowed obtaining PLSR models with R2p and RPDp superior to 0.71 and 2.27, respectively, for all PP techniques and attributes evaluated. The most important wavelengths were well distributed within the three operating ranges of the spectroradiometer (Vis-NIR-SWIR). Thus, the methodology proposed by this research was able to simultaneously determine all the three attributes (alkaloids, sugars and yield) with excellent predictive capacity. This is a promising result for genetic improvement and processing of tobacco (as well as other crops), since it is necessary to evaluate a large number of samples within a short period and at a low cost.

PURPOSE: Several treatment options are proposed for the management of pelvic floor myofascial pain (PFMP). Manual therapy, such as vaginal stretching (VS), is one of these options. Photobiomodulation therapy (PBMT) with a laser device is a treatment option for PFMP that has been tested on other muscles. The aim of this study was to evaluate the effect of VS combined or not with PBMT for PFMP treatment. METHODS: One hundred three women with PFMP were enrolled in a double-blind randomized trial and assigned to VS+PBMT (10 treatments over 2 weeks with 100 mw delivering 12 joules to surface intravaginally, using near-infrared light 808 nm) and VS+shamPBMT treatment groups. Pain severity was assessed by Visual Analog Scale (VAS). Pelvic floor muscle function was assessed by Oxford Scale and surface electromyography. Urinary symptoms were evaluated by ICIQ-OAB and ICIQ-SF questionnaires, and intestinal constipation was assessed by ROMA criteria. RESULTS: There was a significant improvement in pain intensity (VAS) after treatment in both groups, with no difference between groups (p = 0.46). More than 50% of the women complained of severe pain before treatment, and after treatments, it was reported by less than 20% of women (p < 0.001), with no difference between groups (p = 0.08). Urinary symptoms improved in both groups (p < 0.001) with no difference between groups (p = 0.37). Intestinal constipation improved in the VS+PBMT group only (p = 0.01). CONCLUSION: VS and VS with near-infrared vaginal laser therapy were equally effective at decreasing myofascial pelvic pain and reducing urinary symptoms TRIAL REGISTRATION: REBEC (Registro Brasileiro de Ensaios Clínicos; Brazilian Registry of Clinical Trials) under no.RBR-2TDCQ4 (November 11, 2018).

BACKGROUND: Central giant cell granulomas (CGCG) of the jaws are osteolytic lesions that may behave aggressively and respond poorly to surgery. Microscopically, in addition to giant cells, there is a mononuclear cell population composed of macrophage/monocytic cells and spindle-shaped cells of mesenchymal origin. Seventy two percent of these tumours harbour mutually exclusive TRPV4, KRAS and FGFR1 mutations. We aimed to assess the mutational status of mononuclear and giant cells and the osteogenic potential of stromal cells in vitro and in vivo. METHODS AND RESULTS: We screened CGCG for signature mutations and used laser-capture microdissection to demonstrate that the mutations are restricted to the mononuclear cells. Additionally, we established CGCG primary cell culture and observed that the cells retained the mutations throughout passages. By flow cytometry, we observed predominance of CD14- CD51- CD61- cells, consistent with the expected profile for stromal cells. Considering the mesenchymal origin of stromal cells, we assessed the osteogenic differentiation potential of CGCG cells in culture by cytochemistry (von Kossa and alizarin red staining), alkaline phosphatase (ALP) activity assay and gene expression of osteogenic markers. CGCG cells presented self-capacity to increase ALP levels in a time-dependent manner and under osteogenic induction presented increasing number of calcium deposits, and overall higher expression of osteocalcin, RUNX2, ALPL and osteopontin than cells without osteogenic induction. A patient-derived xenograft model for CGCG was established, and osteoid material deposition was observed. CONCLUSION: Collectively, the results confirm that the signature mutations are restricted to stromal cells in CGCG, and the in vitro and in vivo results support that these cells have the capacity to differentiate into osteoblasts, in line with the bone formation often observed in the stroma of these lesions.

Método: estudio transversal, cuantitativo con fuentes secundarias de datos sociodemográficos y clínicos de 2.626 gestantes atendidas en una maternidad pública del interior de São Paulo, en 2020. Para el análisis estadístico, se utilizaron las pruebas exactas de Fisher, las pruebas de Mann-Whitney y el modelo de regresión logística. Se consideró estadísticamente significativa una diferencia de p < 0,05. Resultados: la tasa de seropositividad para sífilis entre las gestantes de esta serie fue de 2,74%. Entre los grupos con pruebas positivas y no reactivas, estado civil, ocupación, lugar de residencia y uso de drogas lícitas indicaron diferencias significativas, pero, en el modelo final, solo el estado civil soltero se asoció con las pruebas reactivas (odds ratio: 0,169; intervalo de confianza: 0,04-0,72; y p: 0,016). Conclusión: en este estudio, el estado civil soltero fue el único factor independiente asociado a la serorreactividad para sífilis. Por lo tanto, es necesario crear estrategias dirigidas a mujeres en esta condición, potencialmente reduciendo la tasa de sífilis congénita.


RESUMO Objetivo: identificar a taxa de testes treponêmicos e não treponêmicos reagentes em gestantes no período do parto e analisar os fatores associados a essa sororreatividade. Método: estudo transversal, quantitativo, com fontes secundárias de dados sociodemográficos e clínicos de 2.626 gestantes atendidas em uma maternidade pública do interior paulista, em 2020. Para a análise estatística, foram utilizados os Testes Exato de Fisher, de Mann-Whitney e o modelo de regressão logística. Considerou-se diferença com significância estatística valores de p < 0,05. Resultados: a taxa de soropositividade para sífilis entre gestantes nesta casuística foi de 2,74%. Entre os grupos com testes reagentes e não reagentes à situação maritária, ocupação, local da residência e uso de drogas lícitas indicaram diferenças significantes, mas, no modelo final, apenas a situação marital não casada apresentou associação com os testes reagentes (Odds Ratio: 0,169; Intervalo de Confiança: 0,04-0,72; e p: 0,016). Conclusão neste estudo, a situação maritária não casada foi o único fator independente associado à sororreatividade para a sífilis. Por isso, faz-se necessária a criação de estratégias voltadas para mulheres nessa condição, potencialmente reduzindo a taxa de sífilis congênita.

ABSTRACT Objectives: to analyze the evidence available in literature on factors associated with inadequate treatment of syphilis in pregnant women. Methods: an integrative review, carried out in the LILACS, CINAHL, Web of Science, Scopus, PubMed and EMBASE databases, with controlled descriptors therapeutic and prenatal syphilis. Results: nine publications composed the interpretative analysis, in which low education, income and maternal age, temporary lack of medication and HIV infection were associated with inadequate treatment of syphilis during pregnancy, in addition to delay or absence of prenatal care and receiving the 1st dose of penicillin, lack of tests or treatment less than 30 days before childbirth, and partners' low compliance with treatment. Final Considerations: among the main factors associated with inadequate treatment, clinical and sociodemographic aspects stand out, as well as failures in drug dispensing, prescription and monitoring of treatment of pregnant women and their partners by the health system.


RESUMEN Objetivos: analizar la evidencia disponible en la literatura sobre factores asociados al tratamiento inadecuado de la sífilis en gestantes. Métodos: revisión integradora, realizada en las bases de datos LILACS, CINAHL, Web of Science, Scopus, PubMed y EMBASE, con los descriptores controlados sífilis terapéutica embarazada y prenatal. Resultados: nueve publicaciones componían el análisis interpretativo, en el que la baja escolaridad, renta y edad materna, la falta temporal de medicación y la infección por VIH se asociaron con el tratamiento inadecuado de la sífilis durante el embarazo, además de la demora o ausencia de control prenatal y de recibir la 1a dosis de penicilina, falta de exámenes o tratamiento menos de 30 días antes del parto, y baja adherencia al tratamiento por parte de la pareja. Consideraciones Finales: entre los principales factores asociados al tratamiento inadecuado se destacan los aspectos clínicos y sociodemográficos de la gestante, así como las fallas en la dispensación, prescripción y seguimiento del tratamiento de la gestante y su pareja por parte del sistema de salud.

Introdução: o aumento no número de casos de sífilis em gestantes, tendo como consequência a sífilis congênita, indica que a doença persiste e continua desafiando os serviços de saúde. Objetivo: analisar a taxa de testes treponêmicos e não treponêmicos reagentes e não reagentes em gestantes, no período do parto, e os fatores associados a esses resultados. Métodos: estudo transversal, retrospectivo e quantitativo, realizado a partir de fontes secundárias de informações (prontuários eletrônicos), com dados sociodemográficos e clínicos de gestantes atendidas em uma maternidade de Ribeirão Preto, de janeiro a dezembro de 2020. Para investigação da sífilis, foi considerada a triagem com teste rápido treponêmico e teste Venereal Disease Research Laboratory (VDRL) para confirmação diagnóstica de sífilis. Os dados do prontuário eletrônico foram digitados no sistema REDCap para organização do banco de dados e analisados por meio do programa STATA SE, versão 14. Foram utilizados os testes Exato de Fisher (para comparação das variáveis qualitativas com a variável dependente) e de Mann-Whitney (para análise de associação entre variáveis quantitativas). A análise dos fatores associados à sororreatividade dos testes treponêmicos e não treponêmicos foi realizada utilizando o modelo de regressão logística. Foram inseridas no modelo final as variáveis que apresentaram na análise univariada p valor <0,20. Para todas as análises foi adotado nível de significância α=0,05. Resultados: os dados avaliados foram de dois grupos de gestantes, sendo o grupo reagente (gestantes que apresentaram testes treponêmicos e não treponêmicos reagentes) e o grupo não reagente (gestantes que apresentaram testes treponêmicos não reagentes). Foram incluídas 2.626 parturientes. Dessas, 2.554 (97,26%) eram do grupo não reagente e 72 (2,74%) do grupo reagente, com uma taxa de positividade de 2,74%. As variáveis situação maritária (p=0,000), ocupação (p=0,019), local da residência (p=0,012) e uso de drogas lícitas (p=0,035) apresentaram significância estatística entre os dois grupos. Por meio da regressão logística, a situação maritária - OR (0,169), IC (0,04-0,72), p=0,016 - foi apontada como associada à ocorrência de gestantes que chegam à maternidade com testes treponêmicos e não treponêmicos reagentes, ou seja, ser casada é um fator protetor contra a sífilis, porém isso não isenta a investigação da doença no parceiro. Dos recém-nascidos (RN) das 72 gestantes do grupo reagente, 42 apresentaram teste de VDRL realizado no sangue com resultados reagentes e 25 que realizaram o exame no liquor apresentaram resultados não reagentes. Dos 42 RN com resultados reagentes, 17 receberam tratamento com penicilina. Além disso, outros 4 RN foram tratados, mesmo o RN tendo o exame negativo. Conclusão: variáveis sociodemográficas influenciam a taxa de sororreatividade de pacientes que chegam à maternidade com sorologia para sífilis reagente, sendo que a situação maritária não casada foi apontada neste estudo como fator isolado e independente à sororreatividade para a sífilis. Portanto, visando à eliminação da sífilis congênita, faz-se necessária a criação de melhores estratégias voltadas para mulheres nesta condição


Background: the increase in syphilis cases numbers in pregnant women, resulting in congenital syphilis, indicates that the disease persists as a challenge to health services. Objective: to identify the positivity rate and the factors associated with reactive treponemal and non-treponemal tests in pregnant women at delivery. Methods: a cross-sectional and retrospective study, quantitative carried out from secondary sources of information (electronic medical records), with sociodemographic and clinical data of pregnant women attended at a maternity hospital in Ribeirão Preto, SP, Brazil, from January to December 2020. Screening with rapid treponemal test and Venereal Disease Research Laboratory (VDRL) test was considered for diagnostic confirmation of syphilis. Data from electronic medical records were entered into REDCap system to organize the database and analyzed using the program STATA SE, version 14. Fisher's exact test was used to compare qualitative variables with dependent variable. We used Mann-Whitney test to analyze association between quantitative variables. The analysis of factors associated with the reactivity of treponemal and non-treponemal tests was performed using logistic regression model. The variables that presented p value < 0.20 in univariate analysis were included in the final model. We used a significance level of α= 0.05 for all analyses. Results: we evaluated data from two groups of pregnant women, one is the reagent group (pregnant women who presented reactive treponemal and non-treponemal tests) and the non-reactive group (pregnant women who presented non-reactive treponemal tests). We included 2,626 parturients. Of these, 2,554 (97.26%) belonged to non-reactive group and 72 (2.74%) to reactive group, with a prevalence of 2.74%. Some qualitative items such as race and pregnancy outcome also did not show significance. Regarding marital status (p=0.000), occupation (p=0.019), residence (p=0.012), use of licit drugs (p=0.035), these variables were statistically significant between the two groups. Through logistic regression, the marital situation without a partner - OR (0.169), CI (0.04 0.72), p=0.016 - was identified as associated with the occurrence of pregnant women arriving at the maternity hospital with treponemal tests and non-treponemal reagents, however this does not eliminate the investigation of the disease in the partner. Of the newborns (NB) of the 72 pregnant women in the reagent group, 42 had a VDRL blood test with reactive results and 25 who underwent the liquor exam had non-reactive results. Of the 42 NB with positive results, 17 received treatment with penicillin. In addition, other 4 NB were treated, even though the NB had a negative test. Conclusion: sociodemographic variables influence the positivity rate of patients who arrive at the maternity hospital with positive syphilis serology, and the marital situation, without a partner, was pointed out in this study as an isolated and independent factor for this positivity. Therefore, it is necessary to create strategies aimed at women in this situation for congenital syphilis elimination

Resumo Embasado em Axel Honneth, este estudo teórico descreve elementos da teoria do reconhecimento e suas interfaces com a autonomia de usuários(as) no cuidado em saúde, retratando as esferas do direito, do amor e da solidariedade em suas interfaces com a autonomia. A partir de considerações reflexivas sobre a prática profissional e o cuidado ofertado, conclui-se que a teoria do reconhecimento pode servir como estrutura que amplia a compreensão de situações do contexto de saúde, especialmente aquelas em que os elementos da esfera de reconhecimento são suprimidos ou prejudicados. A translação da teoria para o campo do cuidado de saúde abre perspectivas interessantes para compreender o valor do direito, do amor, da solidariedade na saúde e de possíveis consequências de sua inobservância.


Abstract Based on Axel Honneth, this theoretical study describes elements of the theory of recognition and its interfaces with user autonomy in healthcare, describing how the spheres of rights, love, and solidarity intersect with autonomy. From reflections on professional practice and the care offered, one concludes that the theory of recognition can serve as a framework that expands the understanding of healthcare situations, especially those in which elements of recognition are suppressed or impaired. Applying the theory to the field of healthcare opens interesting perspectives for understanding the value of rights, love, and solidarity in healthcare and the possible consequences of their non-observance.


Resumen Basado en Axel Honneth, este estudio teórico describe elementos de la teoría del reconocimiento y sus interfaces con la autonomía de los(las) usuarios(as) en el cuidado en salud, retratando las esferas del derecho, del amor y de la solidaridad en sus interfaces con la autonomía. Con base en las consideraciones reflexivas sobre la práctica profesional y el cuidado ofrecido, se concluye que la teoría del reconocimiento puede servir como un marco que amplíe la comprensión de las situaciones en el contexto sanitario, especialmente aquellas en las que los elementos de la esfera del reconocimiento están suprimidos o perjudicados. La traslación de la teoría al ámbito del cuidado de la salud abre interesantes perspectivas para comprender el valor del derecho, del amor, de la solidaridad en la salud, y las posibles consecuencias de su desconocimiento.

Thirteen female Wistar rats were divided into two groups: one treated with ethanol and the other of untreated. Four newborns from each mother were selected and weighed, measured, and evaluated for physical characteristics. From these neonates, chondrocytes were extracted from the articular cartilages of the femur and tibia, and cultivated in a chondrogenic medium at 37oC and 5% CO2. At 7, 14, and 21 days of cultivation, alkaline phosphatase activity tests, MTT conversion to formazan, and percentage area covered by cells per field were performed. At 21 days, the percentage of PAS+ areas in 3D cultures was performed, as well as the evaluation of gene transcript expression for aggrecan, SOX-9, collagen type II, collagen X, Runx-2, and VEGF by real-time RT-PCR. The means were compared by Student's t-test. The weight of the ethanol group neonates was significantly lower than that of the controls. Chondrocyte cultures from the ethanol group showed significantly higher AP activity, MTT conversion, and cell percentage. There was higher expression of collagen type II and lower expression of SOX-9 in the ethanol group. There was no difference in the percentage of PAS+ areas in pellets and in expression of aggrecan, collagen X, Runx-2, or VEGF between groups. In conclusion, prenatal exposure to ethanol alters the phenotype and activity of offspring chondrocytes, which may be mechanisms by which endochondral bone formation is compromised by maternal ethanol consumption.

OBJECTIVE: The purpose of this study was to assess the prevalence of postural changes in women who had urinary incontinence (UI) with myofascial dysfunction (MD) and women who had UI without MD in the pelvic floor muscles (PFM). METHODS: A cross-sectional study was performed with 234 women who had UI and were at least 18 years old at the urogynecology outpatient clinic of a tertiary academic hospital. The International Consultation on Incontinence Questionnaire-Short Form and the International Consultation on Incontinence Questionnaire-Overactive Bladder were used to collect urinary data. Standing postural assessment was performed using photogrammetry in anterior, posterior, and right and left lateral views and was analyzed with Postural Assessment Software. MD was defined as pain of any intensity during palpation of the PFM, and the strength of these muscles was evaluated using the Modified Oxford Scale. RESULTS: The prevalence of MD in women with UI was 51.7% (121/234). Women with MD had significantly smaller angles in the horizontal alignment of the pelvis in the right-side view (mean [SD] = -11.9 [6.9] degrees and -9.6 [7.1] degrees), left-side view (-13.6 [6] degrees and -11.5 [6.6] degrees), and vertical alignment of the body in the left-side view (3 [1.5] degrees and 3.4 [1.5] degrees), showing anterior pelvic tilt and posterior displacement of the body. CONCLUSION: Women with UI and MD had greater anterior pelvic tilt and posterior displacement of the body than women without dysfunction. IMPACT: This study informs physical therapists and other health care professionals about the prevalence of MD in the pelvic floor muscles of women with UI and highlights the need to rule out MD, because it appears to be a concomitant impairment in women who self-report UI. During a postural screen, health care professionals should look for anterior pelvic tilt relative to horizon when evaluating posture in women with UI and MD. The findings of postural changes in women with MD and UI may influence the PFM assessment. LAY SUMMARY: Women with involuntary urinary loss, pain, and stiffness in the pelvic area may show changes in posture associated with this condition.

This study aimed to evaluate the effect of maternal ethanol consumption during gestation and lactation on bone mass and osteogenic differentiation of mesenchymal stem cells of the bone marrow (BMMSCs) in rats. Thirteen adult Wistar rats were used. The rats were mated, and after confirmation of gestation, (day 0) they were distributed in two groups: the control group and the ethanol-treated group. From the ninth day of gestation, the rats of the ethanol and control groups were administered 40% alcoholic solution (4 g ethanol/kg) and distilled water, respectively, daily via gavage until the thirtieth day of lactation. The BMMSCs were extracted from the right femurs and tibiae and cultured using an osteogenic medium for 7, 14, and 21 days. The conversion of MTT to formazan crystals, alkaline phosphatase activity, and percentages of cells per field were analyzed. The number of mineralized nodules per field was examined, and quantification of the gene transcripts for osteopontin, osteocalcin, and BMP-2 was evaluated on day 21 by real-time RT-PCR. Morphometric evaluations of the percentage of trabecular bone and cortical thickness in the left femur and tibia were performed. The means were compared by the Student's t-test, and the differences were considered significant if p < 0.05. The BMMSCs of the rats that consumed ethanol during gestation and lactation, when subjected to osteogenic differentiation in vitro, demonstrated higher conversion of MTT to formazan, higher alkaline phosphatase activity, a higher percentage of cells per field, higher expression of BMP-2, and higher synthesis of mineralized nodules when compared to those of control rat cells. However, there was no significant difference in the percentage of trabecular bone or cortical thickness between both groups. Hence, the consumption of ethanol during pregnancy and lactation did not alter the trabecular and cortical bone tissues of the femur and tibia compared with that of pregnant and lactating control rats that did not consume alcohol, despite BMMSCs showing higher osteogenic differentiation under in vitro conditions.

OBJECTIVES: This study aimed to evaluate the prevalence and associated factors of pelvic floor myofascial dysfunction (PFMD) in women with urinary incontinence (UI). METHODS: A cross-sectional study was performed. Pelvic floor myofascial dysfunction was defined by pain of any intensity during palpation of the pelvic floor muscles and strength assessed by the modified Oxford scale, and was classified in grade I (mild pain), grade II (moderate), and grade III (severe). Univariate and multivariate analyses were calculated (5% significance level) to seek for the associated factors of PFMD. RESULTS: A total of 234 women with UI were included, 121 women in the PFMD group and 113 in the control group. The mean ages were 52.1 ± 10.2 and 53 ± 8.1 years, respectively. Pelvic floor myofascial dysfunction was more frequent among non-White women, and women with PFMD presented more dyspareunia and recurrent urinary tract infection. There was a higher frequency of mixed UI (MUI; 89.2%) in the PFMD group and stress UI (31.9%) in the control group. The prevalence of PFMD was 51.7%, with 10.6% of patients categorized into grade I, 18.8% in grade II, and 22.2% in grade III. In women with PFMD, there is an inverse relation between pain and muscle strength, with the highest strength values (3, 4, and 5) being in grade I (P = 0.028). The multivariate analysis has shown that women with MUI had a risk 4.9 times greater (adjusted odds ratio, 4.93 [2.34-10.42]; P < 0.001) of having PFMD, and non-White women had a 2.1-fold increased risk (adjusted odds ratio, 2.07 [1.10-3.91]; P = 0.024) of presenting with PFMD. CONCLUSIONS: A high prevalence of PFMD was found among women with UI. Mixed UI and non-White race were the associated factors of PFMD.
